چکیده

HIF-1a, one of the most extensively studied oncogenes, is activated by a variety of microenvironmental factors. The resulting biological effects are thought to depend on its transcriptional activity. The RNAse enzyme Dicer is frequently downregulated in human cancers, which has been functionally linked to enhanced metastatic properties; however, current knowledge of the upstream mechanisms regulating Dicer is limited. In the present study, we identified Dicer as a HIF-1a–interacting protein in multiple types of cancer cell lines and different human tumors. HIF-1a downregulated Dicer expression by facilitating its ubiquitination by the E3 ligase Parkin, thereby enhancing autophagy-mediated degradation of Dicer, which further suppressed the maturation of known tumor suppressors, such as the microRNA let-7 and microRNA-200b. Consequently, expression of HIF-1a facilitated epithelial-mesenchymal transition (EMT) and metastasis in tumor-bearing mice. Thus, this study uncovered a connection between oncogenic HIF-1a and the tumor-suppressive Dicer. This function of HIF-1a is transcription independent and occurs through previously unrecognized protein interaction–mediated ubiquitination and autophagic proteolysis. Research Article Cell biology

Tumor and Stem Cell Biology The Hypoxia-Associated Factor Switches Cells from HIF-1a– to HIF-2a–Dependent Signaling Promoting Stem Cell Characteristics, Aggressive Tumor Growth and Invasion

Most solid tumors and their metastases experience periods of low oxygen or hypoxia, which is of major clinical significance as it promotes both tumor progression and resistance to therapy. Critical mediators of the hypoxic response are the hypoxia-inducible factors HIF-1a and HIF-2a. The HIFs are nonredundant and regulate both overlapping and unique downstream target genes. Here, we describe a ...

Regulation of tumor angiogenesis by p53-induced degradation of hypoxia-inducible factor 1a

The switch to an angiogenic phenotype is a fundamental determinant of neoplastic growth and tumor progression. We demonstrate that homozygous deletion of the p53 tumor suppressor gene via homologous recombination in a human cancer cell line promotes the neovascularization and growth of tumor xenografts in nude mice. We find that p53 promotes Mdm2-mediated ubiquitination and proteasomal degradat...

Role of HIF-1a in regulating autophagic cell survival during cerebral ischemia reperfusion in rats

Hypoxia-inducible factor-1a (HIF-1a) plays a beneficial role during cerebral ischemia reperfusion (IR), but the underlying molecular mechanisms are not completely understood. Here, we aimed to investigate the effects and molecular regulation of HIF-1a on brain cell apoptosis and autophagy during IR. We found that augmentation of HIF-1a in re-perfused hematopoietic cells significantly reduced br...

Targeted Replacement of Hypoxia-Inducible Factor-1A by a Hypoxia- Inducible Factor-2A Knock-in Allele Promotes Tumor Growth

Hypoxia-inducible factors (HIF) are essential transcriptional regulators that mediate adaptation to hypoxic stress in rapidly growing tissues such as tumors. HIF activity is regulated by hypoxic stabilization of the related HIF-1A and HIF-2A subunits, which are frequently overexpressed in cancer cells. To assess the relative tumor-promoting functions of HIF-1A and HIF-2A directly, we replaced H...
